Дослідження впливу поліетиленгліколів великої молекулярної маси і декількох полоксамерів на цілісність і плинність мембран еритроцитів методом спінових зондів

Анотація

Методом спінових зондів вивчені механізми впливу поліетиленгліколів (ПЕГ) на плинність ліпідів мембран еритроцитів, можливого плавлення ліпідів мембран і порушення цілісності мембран еритроцитів з часом. Введення 15 % ПЕГ-1500 в еритроцити людини і щура показало збільшення плинності ліпідів мембран еритроцитів на 16 або 30 %, але потім плинність мембран зменшувалась внаслідок дегідратації і стиснення клітини. Введення гідрофільних полоксамерів 407 і 338 в еритроцити приводило до плавлення ліпідів мембран (ефект збільшення плинності мембран 50 – 70 %), але цілісність більшості клітин зберігалася. Перед руйнуванням мембран в'язкість цитозолю різко знижувалась, спостерігалася анізотропія спектрів ЕПР спінових зондів, що пов’язано з суттєвими змінами в структурі цитозолю еритроцитів. Залучення до експерименту полоксамерів, що містять етиленглікольні та пропіленглікольні ланки, показало механізми справжнього плавлення мембран з максимальним збільшенням плинності ліпідів (ефект 70 %), що межує з частковим гемолізом і коагуляцією клітин. Плавлення фосфоліпідів мембран еритроцитів під дією полоксамерів 407 і 338 показало, що справжньою причиною плавлення виступають гідрофобні метиленові групи ПЕГ або ланки пропіленгліколю полоксамерів, що руйнують структуру мембран клітин з усіх напрямків.
