Квантовохімічне моделювання взаємодії епірубіцину з фулереном та вуглецевою графеноподібною площиною
Анотація
Створення нових препаратів «адресної доставки» є одним із пріоритетних напрямків фармакології. Особливо це актуально при лікуванні онкологічних захворювань. Лікарські речовини, зокрема антибіотики антрациклінового ряду, іммобілізовані на поверхні нанорозмірних носіїв для адресної доставки ліків до цільових органів або тканин-мішеней, дозволяють створити оптимальну концентрацію лікарського препарату в зоні реалізації терапевтичного ефекту. Такий метод введення лікарських препаратів істотно знижує їхню системну токсичність за рахунок зменшення загальної дози і більш тривалого утримання діючої речовини в осередку ураження, а також сприяє підвищенню розчинності та біодоступності ліків. Одними із перспективних допоміжних речовин (носіїв) є нанорозмірні вуглецеві матеріали, зокрема, фулерени та графени.
На сьогодні особливості взаємодії епірубіцину з графеноподібною площиною (ГП) та фулеренами на атомарному рівні залишаються маловивченими. Тому ми дослідили методами квантової хімії енергетичні параметри взаємодії ГП і фулерену (С60) з епірубіцином в різних протолітичних формах, які існують при різних значеннях рН водного середовища.
Розрахунки виконували за допомогою програми MOPAC2016, використовуючи метод PM6-D3H4X, в якому, крім врахування водневих зв’язків, беруться до уваги також дисперсійні взаємодії.
На основі аналізу результатів квантовохімічних досліджень передбачається термодинамічна ймовірність перебігу процесу адсорбції епірубіцину на ГП та фулерені в усьому інтервалі рН водного середовища, про що свідчать від’ємні значення ентальпій взаємодії в усіх чотирьох випадках. Встановлено, що найвищою буде теплота адсорбції епірубіцину (протонованої форми) як на графеновій площині (-209.1 кДж/моль), так і при взаємодії з молекулою фулерену (-121.3 кДж/моль).
